This research was conducted to determine the impact of high sodium intake on various obesity-related genes. Four week old Sprague-Dawley rats were purchased at Daehan Biolink. After one week of fertilization, the rats were used at 5 weeks of age. Rats were divided into three groups that received sodium orally at 0 mM(CON), 100 mM(Salt-100 mM), or 400 mM(Salt-400 mM). After 3 weeks, rats were sacrificed and used in the experiment. Triglycerides (TG) and total cholesterol (TC) were measured in the blood serum and liver. The activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were analyzed to test the hepatic toxicity. Genes related to lipogenesis and adipogenesis, uncoupling protein1 (UCP1), and tumor necrosis factor-alpha (TNF-α) were expressed in the liver, white adipose tissue (WAT) and brown adipose tissue (BAT). The levels of albumin and creatinine were measured in the urine. The TG and TC levels in blood serum were highest in the Salt-100 mM group, while liver TG and TC were highest in the Salt-400 mM group. Lipogenesis and adipogenesis related genes in the liver did not increase proportionally to salt concentration. In visceral and subcutaneous fat, peroxisome proliferator-activated receptor gamma (PPARγ) and CCAAT/enhancer-binding protein alpha (C/EBPα) were upregulated proportionally with increasing sodium levels. These results suggest that high sodium intake provoked adipogenesis in SD rats. In BAT, the expression of UCP1 was lower in the Salt-400 mM group than the CON group. Additionally, TNF-α in visceral fat and urine albumin and creatinine were higher in salt treated groups than the CON group. Overall, the results indicate that high sodium intake may be associated with obesity and complications.
