As a result of searching for natural extracts for UV protection and antioxidant effects, we found that the extract of Sophora Flavescens Root Extract has excellent anti-aging effects. Exposure to UV rays caused by frequent going out causes many adverse effects, intensifying inflammation of the skin, and contributing to the production of free radicals and ROS (Relative oxygen species). Skin protection from UV rays and inhibition of free radicals play an important role in healthy and elastic skin cells. Sophora Flavescens Root Extract has been used as a variety of herbal ingredients and is known for its various effects in anti-aging cosmetics. but little research has been conducted on its UV protection and antioxidant efficacy. In this study, the Sophora Flavescens Root were extracted with ETOH, an organic solvent, and Kurarinone was confirmed by HPLC, and it was proved as a related Kurarinone index component. As a result, it was possible to measure the content of Kurarinone from the extract of Sophora Flavescens Root. As a result of the antioxidant experiment, Sophora Flavescens Root Extract was found to inhibit DPPH, known as a free radical. Also, it was confirmed that human keratinocytes treated simultaneously with ROS (H2O2) and Sophora Flavescens Root Extract increased cell viability. Furthermore, keratinocytes exposed to ultraviolet rays was proliferated, and changes in the expression levels of skin inflammatory proteins such as Cyclooxygenase-2 (COX-2) were inhibited by the extract. These results suggest that Sophora Flavescens Root Extract is a potential candidate material that can be used as an antioxidant or for anti-aging through its UV protection efficacy.
