Recently, Myelophycus simpelx (Harvey) Papenfuss, a brown alga, has been reported to exert various pharmacological effects, but studies on its anti-inflammatory and antioxidant ef- fects do not exist. In this study, we investigated the inhibitory effect of M. simpelx methanol extract (MEMS) on inflammatory and oxidative stress induced by lipopolysaccharide (LPS) in RAW 264.7 murine macrophages. The results indicated that MEMS significantly reduced LPS-in- duced release of proinflammatory mediators, including nitric oxide (NO) and prostaglandin E2, associated with decreased expression of inducible NO synthase and cyclooxygenase-2. MEMS also inhibited the LPS-induced expression of proinflammatory cytokines, including interleukin-6 and tumor necrosis factor-α, thereby reducing their extracellular secretion. In addition, MEMS attenuated LPS-mediated cytosolic translocation of nuclear factor-kappa B (NF-κB) and degradation of the inhibitor of nuclear factor-κB-α, which was associated with the inhibition of Toll-like receptor 4 (TLR4) and myeloid differentiation primary response protein 88 expression. Furthermore, MEMS significantly suppressed LPS-induced reactive oxygen species generation and enhanced the activation of nuclear factor-E2-related factor 2 (Nrf2) and the expression of heme oxygenase-1 (HO-1) in LPS-stimulated RAW 264.7 macrophages. These findings suggest that MEMS exerts its inhibitory effect against LPS-induced inflammatory and antioxidant stress in RAW 264.7 macrophages by targeting the TLR4/NF-κB pathway, which might be mediated by the activation of the Nrf2/HO-1 pathway.
